The role of H2B ubiquitylation in development

چکیده
The formation of a multi-cellular organism requires co-ordinated rounds of cellular proliferation, differentiation and apoptosis, to generate, specify and shape body tissues. Strict regulation of proliferation and apoptosis are important for preventing uncontrolled cell growth, the hallmark of cancer. Meanwhile, spatial and temporal control of gene expression gives rise to a myriad of specialized cells from a single genome. Such differential expression patterns are initially established through transient signals, but long-lasting, epigenetic changes are required to maintain these patterns throughout the cell lineage. Amongst these changes is the reversible addition of a single ubiquitin moiety to a specific lysine residue of histone H2B. Ubiquitylated H2B (H2Bub1) is a requirement for diand tri-methylation of lysines 4 and 79 of histone H3 (H3K4me2/3 and H3K79me3), but its biological effects extend beyond downstream regulation of these

H2B ubiquitylation plays a role in nucleosome dynamics during transcription elongation.
The monoubiquitylation of histone H2B has been associated with transcription initiation and elongation, but its role in these processes is poorly understood. We report that H2B ubiquitylation is required for efficient reassembly of nucleosomes during RNA polymerase II (Pol II)-mediated transcription elongation in yeast. This role is carried out in cooperation with the histone chaperone Spt16, a...
متن کاملUbiquitylation of histone H2B controls RNA polymerase II transcription elongation independently of histone H3 methylation.
Transcription by RNA polymerase II (polII) is accompanied by dramatic changes in chromatin structure. Numerous enzymatic activities contribute to these changes, including ATP-dependent nucleosome remodeling enzymes and histone modifying enzymes. Recent studies in budding yeast document a histone modification pathway associated with polII transcription, whereby ubiquitylation of histone H2B lead...
متن کاملTranscriptional activation via sequential histone H2B ubiquitylation and deubiquitylation, mediated by SAGA-associated Ubp8.
Gene activation and repression regulated by acetylation and deacetylation represent a paradigm for the function of histone modifications. We provide evidence that, in contrast, histone H2B monoubiquitylation and its deubiquitylation are both involved in gene activation. Substitution of the H2B ubiquitylation site at Lys 123 (K123) lowered transcription of certain genes regulated by the acetylat...
متن کاملRAD6-Mediated Transcription-Coupled H2B Ubiquitylation Directly Stimulates H3K4 Methylation in Human Cells
H2B ubiquitylation has been implicated in active transcription but is not well understood in mammalian cells. Beyond earlier identification of hBRE1 as the E3 ligase for H2B ubiquitylation in human cells, we now show (1) that hRAD6 serves as the cognate E2-conjugating enzyme; (2) that hRAD6, through direct interaction with hPAF-bound hBRE1, is recruited to transcribed genes and ubiquitylates ch...
متن کاملHistone H2B ubiquitylation and H3 lysine 4 methylation prevent ectopic silencing of euchromatic loci important for the cellular response to heat
In Saccharomyces cerevisiae, ubiquitylation of histone H2B signals methylation of histone H3 at lysine residues 4 (K4) and 79. These modifications occur at active genes but are believed to stabilize silent chromatin by limiting movement of silencing proteins away from heterochromatin domains. In the course of studying atypical phenotypes associated with loss of H2B ubiquitylation/H3K4 methylati...
متن کاملHistone H2B ubiquitylation is associated with elongating RNA polymerase II.
Rad6-mediated ubiquitylation of histone H2B at lysine 123 has been linked to transcriptional activation and the regulation of lysine methylation on histone H3. However, how Rad6 and H2B ubiquitylation contribute to the transcription and histone methylation processes is poorly understood. Here, we show that the Paf1 transcription elongation complex and the E3 ligase for Rad6, Bre1, mediate an as...
